Any ideas on what guitar setup (guitar-amp-effects) to get a simmilar
sound to the melody lead guitar on The Strokes' Reptilia. It sounds
really fat and tight.

Oooh, me sir, me sir - according to my recollection of the only edition
of Guitarist I have, they use a 1985 Jap Strat, a 335 of some
description, and Fender Blues DeVille amps. Didn't recall any effects
pedals being mentioned, and they're rather perfectionist about their
sound.

Or achingly-trendy-anally-retentive depending on what you think of them
:->

The 335 has P90s, and they both use Jeckall & Hyde pedals live. Don't
know what they used on the CD though.

Ok for the Strat, the archtop it's an Epiphone Riviera with P94's single
coils!...
Here's an interesting interview with Nick Valensi:

Great song, we cover this in my band, i use a line 6 flextone amp with all
different amp and cab simulations and can near enough recreate it along with
my les paul, to my ears anyway, i know this is no use though :-) I love
playing this song, especially the bit where everyone stops and its just lead
then the drums/bass come in, then the solo...
